Use of microtuber is advantageous than microplants but dormancy problem of microtuber limits its production and use in the laboratory and the field respectively. This book provides a proven solution of dormancy breaking of freshly harvested microtuber with 5 to 7 days. A series of experiments in relation to production and breaking of dormancy of potato microtubers were conducted during 2000-2003. Nitrogen at 45 and 60 mM with 300-500 mg/l CCC or 30-50 mg/l coumarin or 5 mg/l BAP or 10% sucrose produced higher proportion of large size (>200 mg) microtubers after 105 days. Treatment of intact microtuber with GA3 (0, 5, 10 and 15 mg/l) did not show any noticeable effect on dormancy breaking, while soaking of peeled microtubers treated with 0.1-1.0 mg/l GA3, 0.1 to 1.0 mg/l IAA and 0.01 to 0.1 mg/l Kin for 1 to 25 min and put on water soaked cotton pad containing in petriplates showed >90% sprouting within 7 days.
